During the past two decades, a plethora of studies pointed to what have been described as leukemic stem cells (LSCs), with distinctive role in initiation, maintenance, and relapse of acute myeloid leukemia (AML) due to the resistance of these cells to treatments.

these LSCs are mostly enriched in CD34+CD38- cell population, hence, the assay, isolation, and in vitro culture of this population to study its properties and response to treatment became required to determine the most effective protocols in treating leukemic patients.

Based on that, we measured CD34+CD38- cells in a group of newly diagnosed untreated AML patients, then we sorted them using flow cytometry sorter followed by culturing them to study their proliferative behavior in comparison to CD34+CD38+ cells.

Additionally, we compared the proliferative behavior of mononuclear cells with that of CD34+ cells purified using a positive selection method.

our results were consistent with the presence of LSCs in a higher proportion within CD34+ population, particularly in CD34+CD38- population whose proliferation was slower than CD34+CD38+, an expected finding due to the quiescence characteristic of LSCs.

we believe that we established in this research a concrete basis for isolation and culturing of lscs in preparation for applying drug combinations targeting these cells mainly accused of resisting traditional chemotherapies and causing relapse.
