Acute renal failure (ARF) is the most life-threatening complication of rhabdomyolysis.

Glycerol is commonly used for causing this type of nephropathy in laboratory animals.

This research aimed to investigate the role of Pioglitazone in preventing glycerol-induced acute renal failure, and compare the expected benefit when the drug is administered during two different treatment periods.

24 male Albino Wistar rats were randomly divided into 4 groups (6 animals / group): (A) control, (B) Glycerol50% (10 ml/kg), (C) Glycerol + Pioglitazone (10 mg/kg, for 3 days), (D) Glycerol + Pioglitazone (10 ml/kg, for 6 days).

Pioglitazone was orally intubated, then Glycerol was intramuscularly injected, 24 hours before blood samples collection.

Serum urea (BUN) and creatinine (Scr) levels were measured to assess renal function.

The results showed that BUN and Scr levels were significantly increased in glycerol-injected rats (group B) compared to control group.

While these changes were significantly decreased, in rats treated with pioglitazone for 3days (group C), and for 6 days (group D).

No statistically significant difference was found between the two preventive groups.

Pioglitazone was able to protect the kidney function in glycerol- induced acute renal failure.

This prevention occurred 3 days after Pioglitazone administration; doubling the drug administration period didn't produce any superior benefit.
