Breast cancer is the most common cancer in the world and Arab land, and is responsible for most of cancer deaths of women around the world.

Genetic factors contribute to cancer risk.

One of the genes that affect cancer risk is P53 gene, as it is found to be mutated in about 50% of cancers in total.

R72P polymorphism has the highest frequency of p53 polymorphisms and there are a lot of studies that associate p53R72P with cancer.

The study aimed of to investigate the association between p53 R72P polymorphism and the genetic susceptibility to breast cancer in Syrian women with breast cancer.

The study included 100 women, 70 of them suffer from breast cancer, while 30 of them were healthy.

Both groups were genotyped, for single nucleotide polymorphism, by polymerase chain reaction, and restriction fragment length polymorphism analysis by using BstUI enzyme.

The frequencies of GG, GC and CC genotypes (R72P polymorphism) in patients were 48.6%, 21.4% and 30%, respectively.

While, in control groups their frequency was 43.3%, 43.3% and 13.3%, respectively.

This difference in distribution between patients and controls was statistically significant (P=0.048).

GC genotype was associated with the lowest risk of Breast cancer.

Our study shows that there is an association between p53 R72P polymorphism and breast cancer risk in a group of breast cancer Syrian women
