Background and Objective: complete sealing of the root canal system after cleaning and shaping is critical to prevent re-infecting the root and periapical tissues, Therefore; the aim of this study was to Evaluate the apical sealing ability of three resin-based root canal sealers.

Methods and Materials: The sample of this study consists of 36 recently extracted, single-rooted, upper and lower, permanent human teeth.

The teeth were maintained in 10% formalin for 24 hours and transferred to saline solution at the time of use.

The sample was randomly divided into 3 equal groups, with 12 teeth in each according to sealer used, The root canal of each tooth was prepared using ProTaper rotary files and obturated as follows: Group1:lateral condensation -Adseal, Group2: lateral condensation- AHplus, Group3:lateral condensation- EndoRez.

The teeth surfaces were coated with two layers of nail varnish (except apical portion), and immersed in 2% methylene blue solution (37°C, and 100% humidity) for 24 hours.

The analysis of the penetration of methylene blue solution was performed using a stereomicroscope under 20 x magnifications.

Statistical comparisons were made by using ANOVA and Bonferroni testes.

Results: Data were subjected to statistical analysis P-Value<0.05 was considred as significant.

The results of this study showed that no significant difference in apical leakage between Adseal and AHplus but there was a significant difference between (Adseal, AHplus) and EndoRez in resistance to dye penetration.

Conclusions: Within the limitations of this study, type of root canal sealer plays a role in resistance to dye penetration.
