After the development of root canal preparation systems many studies were carried out about cleaning efficiency of root canals .

Aim of this study was to compare between Protaper (PT) and Twisted file (TF) rotary as preparation systems in root canal cleaning.

Materials and methods: The sample consisted of 20 canals of upper premolar of freshly extracted human teeth .After being cleaned and disinfected they were placed in 10% formalin solution.

Then were randomly assigned into two groups.

After preparation with TF and PT, we evaluate the amounts of debris and smear layer at apical, middle and coronal part of each canal under scanning electron microscopy (SEM).

Data were analyzed by means of the Kruskal Wallis test at 95% level of confidence .

Results:The results showed that there were no statistical differences between TF and Protaper on root canal cleaning , as well as the three root canal parts P> 0.05.

Conclusion: Both rotary systems did not clean the root canal well.
