Cutaneous leishmaniasis (CL) is still endemic and an important health problem in many countries of the Eastern Mediterranean Region.

Cutaneous Leishmaniasis (CL) is a group of diseases caused by a protozoan parasite of the genus Leishmania, which infects millions of people worldwide.

Up to date, no ideal treatment for CL has been identified, which requires access to an effective vaccine emphasize the need for vaccine development based on recombinant proteins or DNA vaccine.

Protein disulfide isomerase (PDI) was identified as one of the virulence factors that involve in Leishmania virulence and survival.

In the present work, it was described the identification, cloning, sequencing of pdI-2 gene.

Initially, The gene was amplified from L.tropica genomic DNA by Polymerase chain reaction (PCR) technique using specific primers.

Then it was inserted into the XhoI / XbaI region of the expression vector pCI, under the transcriptional control of the CMV promoter.

The pCI / pdI-2 construction was transformed into E.

coli DH10B.

Finally, cloned gene sequence analysis revealed that, no mutations were introduced during the cloning process.

This work will lead later to study the protective role of this antigen from the experimental infection Leishmania parasite.
