تم استخدام الكالوس (Callus) و المعلق الخلوي (Cell Suspension) من أجل تقييم الاستجابة للإجهاد الملحي في نوعين من سلالات البندورة (JO112) و (JO992).
تمت زراعة الكالوس و المعلق الخلوي في وسط غذائي (Medium) مزود بأملاح (NAC1 و CAC12) بالتراكيز التالية (0، 50، 100، 150، 200) ملمول بنسب مولارية متساوية.
نقص نمو الكالوس، و قد نقص حجم الخلية المضغوط (packed cell volume)، و نمو الخلية (الوزن لطري و الوزن الجاف) بطريقة مضطردة مع زيادة مستوى الملح.
لوحظ حدوث زيادة في تركيز الايونات (NA، C1، CA) في الكالوس مع زيادة الملوحة في كلا السلالتين.
كما زاد تراكم البرولين (proline) مع زيادة الملوحة.
إن زيادة تركيز الملوحة خفضت بشكل ملحوظ الجهد الاسموزي للمحاليل الخلوية داخل الخلايا.
و قد أوضحت النتائج أن السلالة (JO992) بشكل عام كانت أكثر في الوزن الرطب للكالوس، و الوزن الجاف للكالوس، و محتوى البرولين و البروتين من السلالة (JO112).
النتائج التي حصلنا عليها من هذه الدراسة تقترح أن زيادة تراكم العناصر (C1, NA, CA) في الكالوس كانت مرتبطة بالإحلال العالي للملوحة داخل الكالوس على حساب محتواها من العنصر (K) مع زيادة الملوحة.
Abstract EN
The use of callus and cell suspension cultures to evaluate salt stress responses in two (JO112 and JO992) tomato (Lycopersicon esculentum Mill.) genotypes was determined.
Callus and suspension cells were grown on media supplemented with 0, 50, 100,150 and 200 mM NaCl and CaCl2 in equal molar ratios.
Callus growth, packed cell volume and cell growth (FW and DW) were significantly reduced with increasing the salt level.
Na, Cl and Ca concentrations were increased in the callus with increasing salinity in both genotypes.
Proline accumulation was increased with salinity increasing.
Induced salinity significantly reduced cell sap osmotic potential in the callus.
The results obtained in this study suggest that the higher Cl , Na and Ca accumulations found in the calli were related to the higher callus dehydration by salinity.
On the other hand, K content decreased with increasing salinity.
