صممت هذه الدراسة للتحري عن التأثير السمي للمستخلصات الكحولي و المائي (البارد و الحار) لنبات المرير Sonchus oleraceus في داخل الجسم الحي، حيث أجريت دراسة تقييم تأثير المستخلصات الثلاث (في الجسم الحي) في سرطان الغدة اللبنية الفأري، و كانت الجرعة المميتة الوسطية (LD50) للمستخلصات الكحولي و المائي الحار و المائي البارد (5 غم / كغم و 6 غم / غم و 8 غم / كغم من وزن الجسم) على التوالي، و قد أعطيت الجرع العلاجية للمستخلصات الثلاث بالحقن تحت الجلد يوميا و للفترات الزمنية (أسبوعان و ثلاثة أسابيع و شهر واحد).
بينت الدراسة المرضية النسجية تغيرات مرضية في بعض الأعضاء، حيث أظهرت المجموعة المعالجة بالمستخلص الكحولي ترسب شديد لمادة النشوان amyloid في بعض الأعضاء مثل الطحال.
و امتلاء الأوعية الدموية بالخلايا الالتهابية كخلايا العدلات و ارتشاح الخلايا في نسيج الرئة.
أظهر المستخلصين المائي (الحار و البارد) ارتشاحا للخلايا الالتهابية كخلايا وحيدة النواة في نسيج الكلية و الرئة، لقد بين الفحص المجهري تنخر معظم الخلايا السرطانية في نسيج الرئة، إضافة لذلك لوحظ تغيرات نخرية طفيفة في النسيج الطبيعي بعد مرور شهر من العلاج بالمستخلصين المائي (البارد و الحار).
Abstract EN
The in vivo study was designed to evaluate the anticancer effects of these three types of the extracts on the AM-3 (Murine mammary adenocarcinoma).
The LD50 was (5 g / kg, 6 g / kg and 8 g / kg) for ethanol, hot aqueous and cold aqueous extracts respectively.
The therapeutic doses of extracts were daily injected subcutaneously for two weeks, three weeks and four weeks.
The histopathological study revealed pathological changes in some organs; the group that treated with ethanol extract showed sever deposition of myeloid in some organs like spleen.
The blood vessel filled with inflammatory cells as neutrophile and cells infiltration in the lung tissue.
Hot and cold aqueous extracts showed inflammatory mononuclear cells infiltration in the kidney and lung, section with necrosis in most of the cancer cells in the lung tissue; with few necrotic changes in the normal tissue were noticed after one month of treatment by hot and cold aqueous extracts.
