في بحث سابق تم عزل ميكروب البستريلا مالتوسيدا من أرانب مصابة بعدة أماكن بمحافظة الشرقية جمهورية مصر العربية و تصنيفها جينيا باستخدام PCR.
في البحث الحالي استخدمت العترات المعزولة في تحضير لقاح معملي بطريقة تفكيك بروتين الخلايا البكتيرية بواسطة الموجات فوق الصوتية و قد أجريت تجربتان الأولى لتحديد جرعة اللقاح المناسبة لتنشيط الجهاز المناعي للأرانب لتكوين أجسام مضادة قادرة على صد الإصابة التجريبية و قد كانت 4 x 910 / سم و التجربة الثانية للتحقق من فعالية اللقاح المعملي و مقارنته بلقاح متداول تجاريا في الأسواق المصرية.
أثبتت النتائج السيرولوجية أن اللقاح المعملى أستطاع أن يحمى الأرانب بنسبة 100% و أن مستوى الأجسام المناعية المضادة أعلى بنسبة معنوية إحصائيا عن تلك التي قيست في المجموعة المحصنة باللقاح التجاري.
هذا يدل على أن تحضير لقاح بهذه الطريقة ممكن أن يساعد في الوقاية من هذا الوباء في الأرانب.
Abstract EN
Previously Pasteurella multocida was isolated and genotyped by the author from different outbreaks in rabbits at Sharkia Governorate, Egypt.
In this work, development of avaccine was carried out in the laboratory by sonication of the isolated bacteria genotype A, B and D.
Two experiments were conducted for evaluation of the trialed vaccine.
The first one was carried out for determination of the protective dose of the vaccine and the second experiment for testing of its immune competency with commercial vaccine.
Immunogenic response was evaluated serologically by ELISA and protection percentage on challenge against infection.
Results revealed that sonicated vaccine containing 4 x 109 colony forming unit (cfu) / ml provoked high level of immunity.
On comparison with commercial vaccine, geometric mean antibody titre of the prepared vaccine was statistically higheralong 4 weeks post vaccination.
Moreoverit produced 100 % protection against challenge while commercial vaccine revealed 80 % protection.
Unvaccinated control grouphad zero protection.
It is concluded that immunization of rabbits with vaccine prepared by sonication of genotyped field P multocida isolate would besupportive means for avoidance of pasteurollosis in rabbitaries.
