Abstract EN

Micropropagation of GF-677 rootstock was accomplished.

Single cuttings 0.5-1 cm in length with one bud were excised from adult trees of GF-677 (Prunus persica × P.amygdalus), grown in the field under natural conditions at 6 years in Jellen Station/ ACSAD in Daraa.

The cuttings were surface disinfected and cultured on WPM medium, containing a combination of growth regulators with different concentrations (BA at 2.2, 4.4, 6.6, 8.8 mM/ l) each with IBA 1 mM/ l.

For rooting, shoots were transformed to WPM medium containing different concentrations (IBA at 2.46, 4.92, 9.84, 14.76 mM/ l).

WPM medium supplemented with BA (8.8 mM/ l) and IBA (1 mM/ l) showed the best result for proliferation (4.58).

The highest rooting rate was obtained when explants were incubated to WPM medium with 9.84 mM/ l IBA under a 7 day darkness period.

The propagated plants were transferred to pots with a mixture of 2:1 (v/v) peat/perlite for acclimatization and transferred to green house after one month then to field.

Acclimatization percentage was 95%.
