Ziziphora canescens is a species of important medicinal plants in Syria due to its medicinal properties as antibiotic, flavors and spices in various foods.

This plant is important, especially in folk medicine in some areas (Kalamoon) on the one hand, and retreat of its spread which may lead eventually to its extinction on the other hand, so a protocol for rapid micropropagation has been developing by using lateral and apical buds on nutrient media MS supplemented with different types and concentrations of plant growth regulators.

The use of 0.1% mercuric chloride (MgCl2) for two minutes gave a better percentage 80% of living and uncontaminated buds.

The differences were not significant with a number of shoots that formed from adding the combination 0.1 BA + 0.1 IBA (8 shoots) or 0.1 Kin + 0.1 IBA (7.8 shoots) while the number of formed shoots decreased when increase concentration of BA and Kin to 0.3, 0.6 and 1 mg/ l.

Multiplication Increased when repeat of culture on the same medium for 7 months, the number of formed shoots were 14 and 14.3 at the sixth subculture when adding 0.1 mg/ l BA or Kin with the presence of 0.1 mg/ l IBA, respectively, while the number of formed shoots were 10 on MS medium without any additions of growth regulators at seventh subculture.

In the rooting stage the best percentage of rooting was 95% and number of roots was 7.1 on medium supplemented with 1 mg/ l NAA compared with 2 mg/ l IBA with the percentage of rooting was 90% and the number of roots was 5.8 with 1 mg/l IBA.

The percentage of healthy and living plants after stage of acclimatization was 95%.

The results contained in this research is the first study on the micropropagation of Ziziphora canescens.
