KCNJ11 gene variations have been shown to be associated with an increased risk for type 2 diabetes.

Since this association may be explained by defects of insulin secretion from beta cell, we investigated whether patients with risk allele of KCNJ11- rs5219 have a low therapeutic response to sulfonylureas.

This study involved 75 type 2 diabetic patients have treated with sulfonylurea for an average of one year.

The KCNJ11- rs5219 polymorphism was genotyped by Restriction Fragment Length Polymorphism (RFLP).

Secondary sulfonylurea treatment failure defined as an A1C ≥ 8.0% after a year of sulfonylurea treatment.

The frequency of the risk allele K was slightly higher in the group of patients who failed to respond to sulfonylurea than in the control group (41.25% vs.

35.7%, P = 0.487).

The frequency of the KK genotype was higher in the treatment failure group (17.5% vs.

14.3%), as well as the EK genotype (47.5% vs.

42.85%), but the differences were statistically insignificant.

Our results showed that there was no significant association between rs5219 polymorphism of KCNJ11 gene and the therapeutic response to sulfonylurea in patients with type 2.
