أجريت هذه الدراسة للتعرف عن انزيمات البيتا-جلوكوسيديز(3.2.1.2.1 EC) في البندورة، و نظرًا لعدم وجود دراسات سابقة لوصف البيتا-جلوكوسيديزات في البندورة، فقد تم استخدام الذرة كشاهد حيث تم عزل مجموع البروتينات النباتية من أعضاء مختلفة من البندورة أشتال صغيرة، أوراق حديثة، أوراق متقدمة، ثمار خضراء غير ناضجة، ثمار حمراء ناضجة و كذلك من أشتال الذرة النامية في الظلام.
إضافة إلى ذلك، استخدم بيتا-جلوكوسيديز الذرة المركب وراثيا.
و للتعرف على النشاط الانزيمي المحتمل، تم تعرض الهلام (الطبيعي والمحور) إلى MUGLC-4.
و لقد تم تعريض البروتينات الذائبة إلى ترسيب متتابع مع سلفات الامونيا (35 %، 65 %).
و لم يظهر الزيموجرام تفاعلا إيجابيا ل MUGLC-4 إلا مع أوراق البندورة الحديثة في العمر بالمقارنة مع الانزيمين الطبيعي و المركب وراثيا للذرة.
إن النتائج الموصوفة هنا تقترح، و لأول في أنسجة 4- MUGLC إلا مع أوراق البندورة الحديثة في العمر بالمقارنة مع الأنزيمين الطبيعي و المركب و اثيا للذرة.
إن النتئج الموصوفة هنا تقترح، و لأول مرة، وجود بيتا- جلوكوسيديز واحد على الأقل في البندورة.
و بالرغم من عدم وجود دلائل لتفاعل 4- MUGLC-4 في أنسجة البندورة الأخرى، فمن المحتما وجود بيتا-جلوكوسيديزات أخرى، لكنهت قد تستخدم مواد مختلفة للتفاعل.
Abstract EN
This study was conducted to detect β-glucosidase (EC 3.2.1.21) enzymes in tomato (Solanum lycopersicum L.).
While no previous reports are available to describe β-glucosidases in tomato, maize (Zea mays L.) was used as a reference.
Total plant proteins were isolated from different tomato organs (young seedlings, young leaves, old leaves, roots, green immature fruits, red mature fruits) and from maize etiolated seedlings.
Furthermore, maize recombinant β-glucosidase 1 (rGlu1) was used.
Soluble protein fractions were subjected to subsequent 35 % and 65 % ammonium sulfate precipitation.
To detect potential enzymatic activities, gels (native and denaturing) were incubated with 4-methylumbelliferyl ß-D-glucoside (4-MUGlc).
Only tomato young leaf zymogram showed positive 4-MUGlc hydrolysis as compared to maize native and rGlu1.
The herein described results suggest, for the first time, the presence of at least one β-glucosidase in tomato.
Although no signs of 4-MUGlc hydrolysis could be resolved for other tomato tissues, further β-glucosidases are presumably present, however, they may utilize different untested substrates.
