English Abstract

The goal of this study was to determine the occurrence of tem and she beta-lactamases genes (extended-spectrum β-lactamase ESBL) in Escherichia coli O157 : H7 isolated from beef, stool andcttale’s raw milk During a period of eight months (August 2011 to March 2012), a total of 675 samples were collected from 225stool hospitalized children suffering from diarrhea, 225beef samples collected from three regions in carcass (shoulders, abdomen and back) and 225 cttale’s raw milk samples collected from different regions in Basrah city (Al-Ashar, Al-Basrah and Al- Hadi).All specimens were screened for the presence of E.

coli and EHEC by cultured on MAC, EMB and SMAC.

A total of 256 (37.9 %) of suspected E.

coli isolates were obtained : 67 (29.8 %) from beef samples and 134 (59.6 %) from stool samples and 55 (24.4 %) from raw milk samples.

The study showed higher occurrence of E.

coli in stool samples followed by beef and milk.

All suspected E.

coli is olateswere tested biochemically and 193(75.4 %) were confirmed as E.

coli.

E.

coli isolates (193) were screened on SMAC to detect NSFEC.

Twenty nine NSFEC (67.4 %) out of 43 isolates were detected in beef samples, 44(38.9 %) out of 113 isolates were detected in stool samples and 18 (48.6 %) out of 37 isolates were detected in raw milk samples.

Latex agglutination test was used to detect serotype O157 : H7 in non- sorbitol fermenting isolate, found that : 2 (0.9 %) isolates from beef, 1(0.4 %) isolate from stool and 1(0.4 %) isolate from raw milk samples.

Four E.

coli O157 : H7 isolates were tested for their antibiotic resistance against 14 antibiotics by the Kirby-Bauer disk diffusion test.

All the isolates were found to be resistant to at least 7 antibiotics to which they were subjected.

Therefore, all these four isolates were considered to be multidrug resistant.

The E.

coli O157 : H7 β-lactamase-producing isolates(4)were further examined by PCR technique using two pairs of primers to amplify both tem and she genes.

The results revealed that all (100 %) of the isolates were positive for tagine, 2 (50 %) isolates yielded amplification products with she-PCR specific primers and2 (50 %) isolates were identified as carrying both temandshvgenes.
