درست قابلية العزلة المحلية Bacillus thuringiensis على إنتاج البلوارت البروتينية و ذلك بفحصها مجهريا و تبين قدرتها على إنتاج البلوارت البروتينية بعد 48 ساعة من بداية النمو.
درس النمط البلازميدي باستخدام تقنية الترحيل الكهربائي في هلام الاكاروز بتركيز 0.7 % و تبين امتلاكها حزمة بلازميدية واحدة كبيرة ذات وزن جزيئي كبير، درس دور الحزمة البلازميدية في إنتاج البلوارت البروتينية من خلال تجارب الاقتران التي أجريت على العزلة B.
thuringiensis المنتجة للبلورات البروتينية و الحساسة لمضاد الريفاميسين المستخدمة كخلايا واهبة، و العزلة Bacillus spp غير المنتجة للبلورات البروتينية و المقاومة لمضاد الريفاميسين التي اعتبرت خلايا مستلمة.
أظهرت نتائج الاقتران الحصول على مستعمرات مقترنة على وسط الأكار المغذي الحاوي على مضاد الريفاميسين و قادرة على إنتاج البلورات البروتينية مما يشير إلى دور البلازميد في إنتاج هذه الصفة و هذا ما أكدته دراسة النسق البلازميدي لكل من الخلايا الواهبة و المستلمة و المقترنة.
Abstract EN
The ability of crystals protein production was studied in local isolate Bacillus thuringiensis by microscope examination, and the crystals protein production were seen after 48 hour from growth starting, the study of plasmid profile of the isolate using gel electrophoresis technique in 0.7% of agaros gel, revealed that it had one Mega plasmid band, the plasmid role in production of crystals protein was studied by using conjugation experiments, that they applied on the local isolate B.
thuringiensis which was crystals protein producer and Rifamycin sensitive as donor cells, with Bacillus spp.
which didn’t produce crystals protein and Rifamycin resistance as recipient cells.
Conjugation experiments assured the transmission of crystals production trait with Plasmid to the recipient cells, this result confirmed that crystals protein trait was Plasmid bourn.
