تعد الخلايا البلعمية احد أهم الخلايا التي تساهم في مناعة الانسان و ذلك من خلال افراز سلسلة من العوامل الكيماوية و التي تسيطر على الاستجابة المناعية للانسان و السايتوكينات احد أهم هذه العوامل.
تعتمد البلاعم في فسلجتها على العديد من المسارات الخلوية لغرض إنتاج هذه السايتوكينات و يعتبر 3-STAT احد أهم هذه المسارات الخلوية التي تسيطر على الية إنتاج السايتوكينات و عوامل النمو في خلية جسم الانسان.
من المعروف أن الاحياء المجهرية المتواجدة في أمعاء الانسان لها دور كبير في إنضاج الجهاز المناعي للانسان و البروبايوتك هي جزء من الاحياء المجهرية التي تعيش في امعاء الانسان.
البروبايوتك هي بكتريا مفيدة لمناعة الانسان عندما يتم تناولها بشكل منتظم لكن الميكانيكية التي تتبعها لغرض أحداث الفائدة لا زال تقيد الدراسة و البحث كونها غير واضحة و غير مفهومة لهذا السبب أجري هذا البحث للاجابة على السؤال العلمي فيما لوكانت هذه البكتريا تعتمد في اليتها للتحوير المناعي على أسلوب تحوير إنتاج السايوكينات من خلال استعمال أسلوب تحوير المسار الخلوي 3-STAT.
أظهرت النتائج لهذا البحث بانبكتريا LCS و هي احد أهم افراد عائلة البروبايوتك قابليتها في السيطرة على إنتاج السايوكينات من خلال استحثاث المسار الخلوي 3-STAT في الخلايا البلعملية المستحثة بفعل TNF.
هذه الدراسة سوف تفتح طريق جديد لاستيراتيجة جديدة في السيطرة على الامراض المناعية و ذلك من خلال السيطرة على المسارات الخلوية باستخدام وسائل مؤمونة كالبروبايوتك بكتريا.
Abstract EN
Macrophages are mononuclear phagocytes generated from monocyte emigrated from blood circulation.
Macrophages mediated the innate and adaptive immunity through different routes, and cytokine production is one of these routes.
Signal transducer and activators of transcription (STATs) are cytoplasmic transcription factors that are key mediators of cytokine and growth factor signalling pathways.
STAT-3 is implicated in macrophage cytokine signalling and production.
It's well reported that the microbiota is very important as it primes the immune system for the antigens encountered later in life.
Probiotics defined as ‘Live microorganisms which when administered in adequate amounts confer a health benefit on the host.
Therefore, the aim of this project was to answer the question whether probiotics induced cytokine production via activation of STAT-3 signalling pathway.
Results showed that probiotic of Lactobacillus casie strain Shirota was successfully induced cytokine production via activation of STAT-3 by anti-inflammatory macrophages induced by TNF-α.
The findings of this study will open new strategy to modulate the immune response by probiotic bacteria leading to treat the diseases that related with irregular cytokine production
