Exposure of spermatozoa lo reactive oxygen species (ROS) has been associated with cellular injury, dial includes DNA damage and lipid peroxidation.
in addition, sperm preparation techniques such as centrifugation are associated with the generation of ROS and an increase in DNA damage therefore, this study aimed at the evaluation of density gradient centrifugation using Isolate, as one of the techniques used in assisted conception centers, by determining the total antioxidant capacity (TAC) of seminal plasma before sperm preparation as well as semen characteristics , ROS and sperm chromatin damage indices in semen samples before and after sperm preparation.
The study included 20 infertile patients with normal and 22PAtients with abnormal semen characteristics.
The control group Consisted of 21 healthy donors, the results revealed that infertile patients with abnormal semen characteristics had significantly lower sperm concentration, motility, total motile sperm (TMS) , and normal sperm morphology than those of the healthy donors.
The infertile patients with normal semen characteristics did not significantly differ from the healthy donors.
An increase in ROS level and sperm chromatin damage indices (COMP X sd AND HGRN) was found in both groups of infertile patients in companion to controls.
Such increase in CQMP.
X and SD were of significant value in infertile patients with abnormal semen characteristics.
The ROS-TAC score was significantly lower in both groups of infertile patients than that of healthy donors.
Significant negative Correlation was also found between sperm chromatin damage indices and sperm concentration motility, TMS in the three studied groups.
After density gradient centrifugation using isolate, .there was a significant improvement in sperm motility, TMS and sperm motion characteristics in all groups.
Also.
ROS levels were decreased significantly in both groups of infertile patients and negatively correlated with sperm concentration and TMS the decrease in COMP and X was of significant value only in donors.
While the HGRN value was decreased significantly in donors and patients.
In conclusion, the density gradient c centrifugation using isolate ensures a comparatively acceptable recovery (average 40%) of spermatozoa with good motility and movement characteristics In addition; it has the advantage of being comparatively rapid and consequently low levels of both ROS generation and DNA damage.
Therefore this technique is ideal for cry preserved samples and for retrograde ejaculation, ooligozoospermic and asthenozoospermic patients.
