English Abstract

Background: Follicular Stimulation Hormone (FSH) is essential for normal gonadal function and it is action is mediated by FSH receptor (FSHR).

Single nucleotide polymorphisms (SNPs) of FSHR determine the ovarian response.

Two SNPs of FSHR gene were identified in most previous studies which showed no significant difference between them and concentrated in one SNPs at codon 680.

Objective: The aim of the study is to investigate the association between polymorphism of follicle-stimulating hormone receptor gene at codon 680 and ovarian stimulation in Palestinian women undergoing in vitro fertilization (IVF).

Material and methods: 60 patients under 35 years old underwent IVF operation were investigated and FSHR Polymorphism at codon 680 were examined by polymerase chain reaction restriction fragment length ( PCR RELF).

Results: It was found that non of patient had homozygous polymorphism Serine/Serine from all sample population and heterozygous type was found 75% and in wild type was 25%.

According to the p.N680S FSHR, it was found that there is no statistically significant difference in basal FSH and basal estradiol level (p-value= 0.40) (p-value= 0.624) respectively, while basal LH was statistically significant difference between the wild type (NN) and heterozygous (NS) (p-value= 0.050).

After ovarian stimulation hormone was used, results illustrated that no statistically significant difference between the wild type (NN) and heterozygous (NS) in the dosage of FSH (p-value= 0.273% ), doses LH (p-value= 0.316%), E2 on day of HCG administration(p-value= 0.057), oocytes numbers that stimulated(p-value= 0.150), and the number of mature oocytes (p-value= 0.115%)in women who underwent the IVF procedure in Al Basma Clinic.

Our results illustrated that the PCO patients had no statistically significant difference in FSH dose of the wild type and heterozygous type(p-value= 0.947).

But was statistically significant difference in the production of oocytes(p-value= 0.008), and in the AMH level between the patients with PCO and patients without PCO (pvalue= 0.021).

Conclusions: FSH receptor polymorphisms for heterozygous (NS) and wild type (NN) are not correlated with ovarian response to ovarian stimulation hormone.

We recommend increasing the numbers of samples.
