Background & Objective: The protein p53 is the guardian of the cell gene contents from changes, and the gene that produces it is called Tumor Suppressor Gene which mutates in 50% of Squamous cell carcinoma’s cases producing defective protein .Studies have not detailed the expression ratio of the defective protein Within the cancer cells, also studies have less details about the cases which did not show a mutated protein.
Investigating the non mutant protein p53 (wtp53) in Squamous cell carcinoma’s samples of the head and neck , and investigating the expression of the proteins ATM and MDM2 and their role in inhibiting the non mutant protein p53 (wtp53) in cancer cells.
Materials & Methods: The sample consisted of a cancer samples included thirty biopsies of squamous cell carcinoma (3 lip,12 Pharynx, 15 oral cavity), and a control sample of five biopsies from healthy free gums.

we made two slices from each sample and stain them double staining Immunohistochemistry, we stained one Slide by two antibodies ATM and wtp53, and the Other slide by two antibodies MDM2 and wtp53.
The statistical experiments were :Person, k2 , phi ,at p<0,05 .
Results: Positive expression for the three proteins appeared in all biopsies slices, and the over-expression proportion of wtP53 and MDM2 and ATM protein in a SCC (42,1%, 51,9%, 59,6%, respectively) .
direct relation between wtp53 and each Of MDM2, ATM was shown in control samples while it was reverse relation in the cancer samples.
Conclusions: The expression of non mutant protein p53 (wtp53) observed strongly in proliferating cells .
And the expression of each of wtp53 and MDM2 and ATM are higher in cancer cells than non mutant one .
while it isn’t effective in cancer cells.
Each of MDM2 and ATM has a role in inactivation wtp53 in the case of cancer.
