An a new digestion method for selenium determination in supplement food with spectrophotometric method using 3,3 Diaminobenzidine Hydrochloride.

New digestion method was used for selenium (IV) extraction from food supplements for direct determination with spectrophotometric method.

The digestion method provides acid digestion of the sample in a closed vessel device using temperature control microwave heating.

After sample digestion and selenium (IV) occur ,spectrophotometry method applied using 3,3 Diaminobenzedine hydrochloride.

Simple, sensitive and specific spectrophotometric method for the determination of selenium at trace levels.

The method based on the formation of stable yellow-colored piazselenol complex by reaction of Se (IV) with 3, 3'-diaminobenzidine hydrochloride, which is then extracted in toluene and detected spectroscopically at 434 nm.

Optimization conditions (pH Effect, thermo effect, time effect, reagent DABH concentration effect extract solvent effect, reagent consequence effect) for piazselenol complex formation were studied.

The microwave digestion method was found to be repeatability and satisfactory recovery from the spiked samples of standard Se (IV) .The spectrophotometric method was validated and found to be suitable for estimation of Se (IV) species in food supplement.
