Arabic Abstract
يوفر توصيل العقاقير عن طريق الجهار الرئوى مزايا متعددة مقارنة بالطرق الأخرى لتوصيل العقاقير تطرا لمساحته الكبيرة و كثافة الأوعية الدموية فيه و توفر عشاء رقيق بين الحويصلات الرئوية و الدم.
يعتبر توصيل العقاقير عن طريق هذا المسار مناسبا للمرضى، فهو يعتبر أقل إيلاما مقارنة بالحق الوريدي او العضلي ويمكن استخدامه لايصال العقار اما للتأثير الموضعي او الكلي.
يتيح استخدام أجهزة الاستنشاق بالتركيبات الجافة ( DPI ) توصيل المكونات الصيدلانية الفعالة ( API ) إلى الرئة بخصائص ملاكمة.
للقيام بذلك، يجب أن تتمتع التركيبة بسمات الجودة الهامة المتعلقة بالحجم و القدرة على التدفق و القدرة على الوصول إلى الأجزاء السفلية من الجهاز التنفسي.
الصياعات التجميعية النانوية هي تقدية مناسبة تمكن من تطوير هذه التركيبات ( DPIs ) بطريقة ناجحة.
لذلك، فإن الهدف من هذا المشروع هو استخدام تكنولوجيا النانو لتطوير جزيئات المحتوية على أكسيد الحديد و المادة الفعالة ( داکتینومايسين ) حيث تقوم جسيمات أكسيد الحديد الثانوي بمثابة حاملة لتوجيه المادة الفعالة للموقع المستهدف في الرئة.
تم إنشاء و تطوير جسيمات أكسيد الحديد الدائوية باستخدام منهجية الجودة حسب التصميم.
كان النموذج الدوائي هو الدكتينومايسين البوليمرات التي تم فحصها هي الكيتوسان و املاح الاجيدات تم التحقق من صحة الطريقة الكمية من قبل كروماتو عراقيا سائلة عالية الضغط ( HPLC ) وفقا لتوجيهات المؤتمر الدولي المعني بالمواءمة تبع ذلك دراسات فحص لتطوير جزيئات أكسيد الحديد النانوية.
تم تمييز جزيئات الدكتينومايسين المنتجة التي تحتوى على جسيمات متناهية الصعر لقطر الأيروديناميكي متوسط الكتلة ( MMAD ) و نسبة الجسيمات الدقيقة ( FPF ), و تأثير اطلاق العقار السريع نسبة الجسيمات الدقيقة لأكسيد الحديد ( FPF ) و الجرعة المنبعثة في دراسات الفحص الأولية لتحديد اهم المدخلات و العمليات الأكثر ملاءمة لدراسة التحصين عن طريق توطيف مبادئ QbD من خلال تصميم التجربة كشفت النتائج تفوق مجاميع الجسيمات الثانوية التي تحتوى على DPI في تقديم جر عة عالية الانبعاث، و ارتفاع نسبة الجسيمات الدقيقة FPF و MMAD المستهدفة.
أظهرت مساحة التصميم في تحليل QbD أنه عند استخدام تركيز الداكتينومايسين بين %4% إلى 5 مصاحبا لتركيز البوليمر يتراوح من 0.5% إلى 0.8% باستخدام الجينات الصوديوم أو تركيز الداكتينومايسين ما بين 2.7 إلى 4% المصاحب لتركيز البوليمر تتراوح بين 1.3 % إلى 2 % باستخدام الجينات الصوديوم سوف تحصل على النتيجة المرجوة.
وبالمثل، تم تحقيق نتائج ملائمة ومستهدفة مع الكيتوران باعتباره بوليمر، عندما كان تركيز الداكتيدومايسين بين 4.5% إلى 5% وتركيز البوليمر تتراوح بين 0.5% الى 0.8%.
لذلك، يمكن أن تكون نتيجة هذا المشروع البحثي نقطة انطلاق لمزيد من العمل لتحسين وتقييم DPI لتقديم عقاقير أخرى التي تستخدم مجاميع الدانو لأكسيد الحديد.
English Abstract
The pulmonary route for drug delivery offers multiple advantages over other routes of drug administration owing to its large surface area, high vascularization and thin blood-alveolar barrier.
Drug delivery by this route is convenient to patients, less painful (non-invasive) and can be employed for local and systemic delivery of active pharmaceutical ingredients (APIs).
The use of dry powder inhalers (DPI) enables the delivery of APIs to the lung with favorable properties.
To do that, powder should possess critical quality attributes pertinent to size, flowability and ability to reach the lower parts of the respiratory system.
Nano aggregate formulations are suitable technology that enables the development of successful DPIs.
Therefore, the aim of this project is to employ Nano technology to develop iron oxide containing nanoparticles using model API- dactinomycin.
Iron oxide nanoparticles will serve as a carrier for directing the API to the targeted site of action within the lung.
The development and optimization of iron oxide nanoparticles was carried out employing Quality by Design (QbD) methodology.
Nanoparticles were built up from iron oxide that was chemically prepared using bottom-up method.
Two polymers were investigated (chitosan and sodium alginate).
The quantitative method for the analysis of dactinomycin by HPLC was validated according ICH guidelines.
That was followed by screening studies for iron oxide nanoparticles development.
The produced dactinomycin containing nanoparticles was characterized for mass median aerodynamic diameter (MMAD), fine particle fraction (FPF), burst effect, iron oxide FPF and the emitted dose in the initial screening studies to investigate the most suitable input and process parameters to take to an optimization study by employing QbD principles through Design of Experiment.
Results revealed the superiority of the nanoparticle aggregates containing DPIs in delivering high emitted dose, high FPF and targeted MMAD.
Design space in QbD analysis showed that when using a concentration of API between 4% to 5% w/w accompanying with polymer concentration ranging from 0.5% to 0.8% w/w using sodium alginate or concentration of API is between 2.7% to 4% w/w accompanying with polymer concentration ranging from 1.2% to 2% using sodium alginate will get the desired outcome.
Similarly, favorable critical quality attributes (CQA) results were attained with chitosan as a polymer, when API concentration was between 4.5% to 5% w/w and polymer concentration ranging from 0.5% to 0.8% w/w.
Therefore, the outcome of this research project could be a starting point for further work to optimize and assess DPI for delivering other drugs employing iron oxide- polymer nano aggregates.
