The main method for removing phenols from the environment include its biotransformation by bacteria in water and soil.
Rhizobium nepotum was able of utilizing phenol as substrate and being sole carbon and energy sources.
Six different trials of phenol concentrations (200, 400, 700, 800, 1000 and 1100 ppm) were initially used.
The phenol was estimated at OD of 510 nm on the base of the condensed phenol with 4- aminoantipyrine, which was followed by the oxidation process with the alkaline potassium ferry cyanide, which gave red color.
Thus, the amount of phenol removal by bacteria can be estimated using the standard curve of phenol and according to the concentration range to be determined.
It was observed that increasing the phenol concentration would cause the phenol degradation rate to boost up to the rate of 6.6 ppm/h for 700ppm accompanied with the highest percentage removal (44%), and then it started to decline with increasing the concentration of phenol.
When using 700 ppm it was perceived that the bacteria were able completely to deteriorate the phenol within about 86 hours of incubation and were parallel with the growth level of the bacteria based on the degree of turbidity.
The best rate obtained was with 28°C (24.1 ppm/h) whereas increasing or decreasing the temperature caused the inhibition in biodegradation rate of phenol.
The optimum pH level for bacterial growth and removal of phenol was 8.
