Role of tumor necrosis factor-alpha on hepatic regeneration after partial hepatectomy in adult male albino rats

Objective : This study aimed to evaluate the effect of TNF- a on liver regeneration after seventy percent partial hepatectomy of male rats.

Methodology : Rals were divided into three groups; control, untreated partially hepnleetomisod group and Penfoxifylline (a drug which blocks TNF-a) treated hepatectomised group, Each group had thirteen rats.

Rats were killed on the 1st, 3rd, 5th and 7th days post hepatectomy.

The removed part of the liver was weighed to detect the increase of the total liver weight on the previous clays.

A sample of the liver was taken for hisiological and immunohistochernical studies.

Results : The weight of liver in the untreated group showed gradual increase up to more than ninety nine per cent of the original liver weight on the 7th day posthepateclomy In the treated group, it reached only fifty five per cent of the original liver weight on the7th day posthepatectomy.

Miiotie activity in (he untreated group was sharply increased on the 1st day then gradually decreased and disappeared on the 7th day.

Treated group showed a decreasing in mitotic activily from (heist day and disappearing on the 3rd day.

Immunohistochemical studies in untreated group showed intense cytoplnsmic ovcrexpression for TNF- a on the 1st and the 3rd days, moderate ovcrexpression on the 5th day and mild ovcrexpression on the 7th day.

Treated group showed mild ovcrexpression on the 1st day and no expression on other day posthepateclomy .Conclusion : The study demonstrated that TNF- a promoted the regenerative changes that occur in the liver after partial hepatectomy.
