Effect of age, duration of infertility, and type of infertility on seminal fiuid analysis paprameters, plasma membrane integrity, and pregnancy rate for infertile patients after intra-uterine insemination technique

This study was designed to evaluate the effect of age, duration of infertility, and type of infertility on seminal fluid parameters, hypo-osmotic swelling (HOS) test, and pregnancy rate for infertile patients undergoing intra-uterine insemination (IUI) techniques.

One hundred infertile couples were enrolled in this study and according into age, duration of infertility, and type of infertility were divided into 5 groups, 6 groups, and 2 groups ; respectively, and semen samples were collected by masturbation and evaluated before and after in vitro sperm activation.

The spermatozoa prepared by simple layering technique by using 1ml of the liquefied semen were layered beneath 1ml of IVF culture medium.

The HOS-test was performed after and before in vitro sperm preparation by mixing 0.1 ml of semen with 1.0 ml of a 150 mOsm / L NaCl as a hypo-osmotic solution.

The sperm function tests (SFTs) including sperm concentration, sperm motility (%), progressive sperm motility (%), and normal sperm morphology (%) were evaluated according to standard World Health Organization (WHO) criteria.

For IUI, the sperm prepared and incubated for 30 minute in 5 % C02 at 37°C after in vitro sperm preparation.

The results of the present study demonstrated that SFA parameters, HOS-test scores, and IUI outcomes decreased with advancing age and duration of infertility.

In addition, the results of SFA parameters and HOS-test for infertile couples with primary infertility were more deviated from normal range of criteria of WHOM than the secondary infertility.

Finally, it was concluded that the age partners, duration of infertility, and type of infertility adversely effect on results of seminal fluid analysis parameters, HOS test, and pregnancy rate of infertile patients after intra-uterine insemination technique.

Further studies are recommended to assess the effect of age, duration of infertility, and type of infertility on pregnancy rates after intra-cytoplasmic sperm injection.
