The protective effect of garlic oil or and thymoquinone against cardiotoxicity of doxorubicin

Doxorubicin is a widely used chemotherapeutic agent for treatment of a variety of human neoplastic diseases.

But its clinical usefulness is hindered by its dose-limiting cardio toxicity.

This cardio toxicity has been suggested to result from the generation of oxygen free radicals.

Garlic oil or thymoquinone were found to inhibit oxidative stress in a wide range of animal models.

This study was designed to assess the cardio protective potential of garlic oil and or thymoquinone against toxicity of doxorubicin using 100 male Sprague Dawely rats divided into 5 groups (20 each): control normal, doxorubicin treated, doxorubicin plus garlic oil, doxorubicin plus thymoquinone, doxorubicin plus garlic oil and thymoquinone.

Serum and cardiac tissue samples were taken.

Development of cardiac damage in doxorubicin treated group was established by increased cardiac content of malondialdehyde (MDA) as well as serum activity of creatine phospo kinase (CPK) and lactate dehydrogenase (LDH).

While, total protein and reduced glutathione (GSH) contents beside activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) were decreased in cardiac tissue of doxorubicin treated group.

Using of garlic oil or thymoquinone during doxorubicin treatment improve all the estimated parameters and so ameliorate the cardiac damage.

In conclusion, administration of garlic oil and / or thymoquinone induce significant improvement in the oxidative status and protect against cardio toxicity of doxorubicin.
