Eighteen populations of three Hyoscyamus species were collected from different ecogeographical sites during Mars-April 2004, across the range of their distribution in Jordan.

In total, 12 populations of H.

aureus, four of H desertorum and two of H reticulatus were collected.

Seventeen different morphological traits and three selected highly polymorphic RAPD primers were used in order to assess the level of genetic variability within and among the collected populations. Polymorphism was common in various degrees in most of morphological traits recorded, which indicates the wide range of variability in most studied traits.

RAPD analysis revealed extensive polymorphism between different genotypes of wild naturally occurring Hyoscyamus populations, with 57 % of the 60 scorable bands showing polymorphism.

Hyoscyamus populations harbor a very high diversity index with a mean of 0.132 range from 0.0734 to 0.1774.

A high overall average of Shannon index of diversity over collected Hyoscyamus populations was obtained, the mean was 0.1992 and a range from 0.1156 to 0.2693.

This range of genetic variability may be due to (i) Very diverse ecogeographical region throughout the kingdom and (ii) High degree of climatic heterogeneity of mountain compared to desert. Based on morphological and diagnostic traits, the cluster analysis of collected populations of the three Hyoscyamus species did not mostly demonstrate any distinct groups within the species.

Using RAPD analysis, most populations of the same species were clustered into groups (as a unique units) except one population of H, aureus, collected from Shoubak, did not belong any group and was distinct in its genetic make-up due to prevailing environmental conditions in Shoubak area.

Results demonstrated that the strong genetic differentiation among species using RAPD as a molecular marker.
