Seven halophile bacterial strains were isolated from three different places in the Dead Sea region, which are AL-Madrasah, AL-Hadeetha and southern east shore of the Dead Sea.

Morphological, biochemical and physiological analyze were done to characterize the seven Isolates.

All of the isolates were Gram negative and different NaCl concentration for growth.

Studying of optimum temperature, optimum pH and optimum salts (NaCl and MgC12), and utilize of organic compounds ; Fructose, Lactose, Glucose, Sucrose, Citrate, Oxalate, Malate, Mannitole, Methionin, Sorbitol, K-Na-Tartarate, Succinate, L-Lysine, Arginine, Na-Acetate, Glycerol, L-Glutamine And hydrolyses of organic compounds as ; casein, gelatin, aesculin, DNA, starch, tween20 and tween80.

The effect of NaCl concentration divided the isolates into three : 1.

Two isolates extreme halophilic bacteria H4 and H5.

They similar to genus Haloferax, especially H.

volcani were isolated from the Dead Sea, with little differences in some characterization.

Four isolates HI, H3, H6 and H7 were moderately halophilic bacteria and similar in morphological, physiological and biochemical characterization.

Many differences between those and known species were isolated from the Dead Sea.

One isolate H2, was moderately halophilic bacteria but difference from other isolates, and some differences from known species were isolated from the Dead Sea.

Identification of halophilic bacteria is not easy especially moderate halophilic bacteria due to high number of them and difficult to get all them.
