Random Amplified Polymorphic DNA (RAPD) markers were performed to fingerprint 14 varieties of peach (Prunus persica L.

Batsch) cultivated in Kurdistan region of Iraq, genetic distances and genetic relationships between these varieties were also studied.

To achieve this, total genomic DNA was isolated from 3gm of fresh leaves.

The average yields of DNA were in the range of 155-995 μg / ml of 3gm fresh tissues with a purity ranging between 1.7-1.8.

DNA polymorphisms were scored within detectable amplified fragments (their numbers and molecular weights) on agarose gel electrophoresis stained by ethidium bromide.

A total number of 36 Operon primers were tested, thirteen primers generated informative RAPD patterns and selected for further RAPD analysis.

These 13 primers yielded 1303 (total numbers) of amplified bands (DNA fragments), out of which 182 were polymorphic bands across tested varieties.

Each selected primer produced between 52 bands (OPB-10) to 205 bands (OPE-20).

Amplification products (DNA amp icons) and fragments size ranged from 202 bp (OPL-20) in cultivar to 4956 bp (OPS-03).

The highest number of polymorphic bands (23 bands) was produced by primer OPE-20 while the lowest number of polymorphic bands (5 bands) was produced by primer OPC-04.

The primers differed in their capacity in producing polymorphic amplified profiles between different peach varieties which individually reflected variety specific DNA profiles (fingerprints).

The primers of most interest of this purpose were those that produced more variety specific DNA profiles, such as OPE-20, OPC-09 and OPG-16.

The efficiency and the discriminatory value of each primer were computed individually.

The primer efficiency ranged from 0.22 (primer OPF-05) to 0.08 (primer OPC-04).

The primer OPE-20 had a highest discriminating power 12.64 % and could fingerprint 9 varieties individually among 14 varieties studied.

The primer OPC-04 had the lowest discriminating power 2.75 % and could not fingerprint any of the varieties Genetic distance by pair wise distance matrices were compiled by the NTSYSPC.

Version 1.8 software estimated among the 14 varieties ranged from 0.2087 between the varieties Yellow coronet and Red mays (the closest varieties in this group) showing 79 % similarity, to 0.6552 between the varieties Yellow coronet and May grand showing just 34 % similarity Cluster analysis (phylogenetic tree) by unweight pair-group method of arithmetic means (UPGMA based dendogram) revealed two main genetic groups (major clusters).

The first genetic group included the following varieties Nectarine 4, Nectarine 6, Yellow coronet, Red mays, Florida sun, springtime, Red leaf (Grafted coronet), Seedy varieties.

The second genetic group included MI ski, Ismailia, Figgy, Abu alma, Silver king, May grand varieties.

The first major genetic group included three subgroups: the first subgroup included Yellow coronet and Red mays, the second subgroup included Florida sun and springtime and the third subgroup includes Red leaf (Grafted coronet) and Seedy.

The second major genetic group included two subgroups: the first subgroup included musky and Ismailia and the second sub-group included Abu Alma and Silver king.

The overall analysis of the results revealed that the genetic relationships between the peach varieties were related to some of their morphological characters as well as to their geographical origins.
