Copper (Cu) is an important cofactor for many enzymes.

It is involved in electron transport, redox and other important reactions.

However, excess of this metal in its ionic form results in cell death.

Moreover, Cu is the most contaminant of industrial effluents; research attention has been focused on biological methods for the treatment of effluents.

For this reason this study is done in order to: characterize the strains of bacteria existing in As-Samra Wastewater Treatment Plant (SWTP), isolate and characterize those able to highly uptake of Cu, also determine the minimum inhibitory concentration (MIC) of Cu and finally determine wither the uptake genes are carried on plasmid or on chromosome. A total of ten (10) species naturally inhabiting SWTP located at Zarqa were isolated, purified and preliminary identified on the basis of morphological, cultural and biochemical characteristics.

These species were examined for their ability to uptake copper ions at concentration of 50 ug / ml from copper nitrate solution and the uptake was found in the following order: E.

coli (410) > B.

cepacia (304) > P.

vulgaris (266) > Acinetobacter (83.6) > Shigella sp.

(37.24) > P.

multocida (34.2) ppm / g wet weight.

K.

oxytoca, V.

fluvialis, C.

freundi and Corynebacterium argentoratense were unable to uptake at the concentration of 50 ug / ml.

Thus E.

coli was selected for the study. Growth of bacterial species is altered by copper ion concentration (20, 40, 50, 60) ppm and inhibited at high concentrations (100, 200, 300), this alteration is mostly remarked on lag phase.

MICs were dependent on the copper compound used (50ppm for copper nitrate trihydrate, 70 ppm for copper sulfate pent hydrate and 100 ppm for copper chloride dehydrate).

MTC of copper sulfate was greater than MIC (105ppm). Examining the effect of increasing concentrations of Cu on the biological uptake revealed that uptake depends on the copper compound used (using copper sulfate gave uptake approximately four times higher than that obtained with copper chloride and copper nitrate, at 600 ppm, uptake was 3443, 1053 and 728 ppm / g wet weight of copper sulfate, copper chloride and copper nitrate respectively).The effect of carbon source on the uptake revealed the order of uptake from high to low : maltose» lactose > sucrose > glucose and that of nitrogen source used was: ammonium chloride < tryptone

coli was 38% reached after 47 h of culture (initial Cu concentration was 40 \ iglm \).

The plasmid profile showed that this E.

coli harbored plasmid, curing of this plasmid was carried out with ethidium bromide and cured cells were able to grow up to 80 ppm of Cu + 2 on solid medium.

Only inhibition of 22 % of uptake was shown in cured cells compared to parental cells 7.5 % of bio sorption and 11.5 % of adsorption.

Thus, uptake and resistant genes may be carried on chromosome.
