This study was performed to evaluate the ability of omega-3 fatty acids (omega-3 FAs) to protect ifosfamide (IF) treated rats against the toxicity of this chemotherapeutic agent.

Thirty adult Wister male albino rats were used in the present study, in which they were randomly divided into six groups (each contained five rats).

These groups included : Group 1: control (given only saline), Group 2: omega-3 (4gm / kg diet), Group 3: IF (50mg / kg b.w.), Group 4: IF (80mg / kg b.w.), Group 5: IF (50mg/kg b.w.) plus omega-3 (4gm / kg diet), Group 6: IF (80mg / kg b.w.) plus omega-3 (4gm / kg diet).The IF was administrated intraperitoneally for five consecutive days, while omega-3 was given orally within the diet for six consecutive days (started the day before IF injection).

Both IF doses caused a significant decrease in the body weight gain of rats when compared with the control rats, while a significant increase in the body weight gain of the rats had been recorded after applying omega-3 to both doses of IF when compared with groups given IF alone.

The biochemical estimation methods showed that IF caused a significant increase in the levels of serum creatinine, phosphorous, glutamate-oxaloacetate transaminase (GOT), glutamate-pyruvate transaminase (GPT) and malondialdehyde (MDA), while it caused a significant decrease in serum glucose and testosterone compared with the control group.

On the other hand, omega-3 FAs had kept all these levels approximately close to the control values when administrated to the IF treated rats.

Both doses of IF significantly decreased WBC, RBC, HGB and PLT when compared with the control group, while omega-3 with IF increased these parameters significantly.

For the histological study, paraffin and plastic methods were employed for the preparation of liver, kidney, cerebrum, cerebellum and testes sections.

This chemotherapeutic agent has induced many histological alterations in the liver such as degeneration of the cells, inflammatory cells infiltration and dilation in the sinusoidal lumen, while it caused severe inflammation, degeneration in the kidney tubule lining cells and hypertrophy of these tubules.

Furthermore, IF has caused degeneration of the pyramidal neurocytes of the 2 nd and 3rd layers of cerebral cortex and the Purkinje cells in the middle layer of cerebellar cortex.

With respect to testes, IF has caused degeneration of the germinal cells lining the testes, in which a large number of the seminiferous tubule lumens appeared empty.

All these organs have been approximately histologically protected from the damaging effect of IF when a combination of omega-3 FAs and IF was administrated.

Interestingly, omega-3 didn’t interfere with the antimitotic property of IF, suggesting a very important role for this oil in the future of cancer chemotherapy.
