الملخص الإنجليزي

The present study was conducted at the animals house of Veterinary Medicine College–University of Basra .

Fourty eight adult local male dogs were used in the present study .The study was divided into two experiments :- First experiments : This experiment was designed to investigate the effects of unilateral adrenalectomy of right adrenal gland on some hematological , biochemical and histological changes for 60 days .

Thereafter to evaluate the role of treatment with DEX and Flud in amelioration the changes that resulted from unilateral adrenalectomy.

24 adult local male dogs weighning 20 – 27 Kg and 2–3 year old were divided into four groups, 6 each as following : First group ( control) Intact non treated animals.

Second group (UADX) : Unilateral adrenalectomized animals.

Third group (UADX-DEX ) unilateral adrenalectomized animals treated orally with tapering doses of Dexamethasone (1,0.5 and 0.25) mg / kg BW / day, for 3 week respectively. Fourth group(UADX-Flud ), unilateral animal treated orally with tapering doses of Fludrocortisone (0.25, 0.12 and 0.06) mg / kg BW /day , for 3 week respectively , all group are leaved until 60 days .

The results revealed the following: 1- A significant (p≤0.05) decrease in the body weight gains, RBC count, Hb concentration and PCV value in the UADX and UADX - Flud treated groups during 8 days of treatments then returned to their normal values compared with control. 2- A significant (p≤0.05) decrease in the total WBC count, lymphocyte and a significant increase in the monocytes , neutrophils and eosinophils percentages were observed in UADX and UADX–Flud treated group during early periods of treatments compared with control . 3- A significant(p≤0.05) decrease in serum T3, T4, cortisol and testosterone and a significant increased in ACTH concentrations were recorded in UADX and UADX –Flud during 15 days of treatment compared with control and UADX –DEX treated groups. 4- A significant (p ≤ 0.05) increase in serum AST, ALT and ALP enzymes concentrations were recorded in all treatments compared with control. 5- A significant(p ≤ 0.05) increase in serum cholesterol and significant (p ≤ 0.05) decrease in serum triglyceride concentration were found in UADX and UADX –Flud compared with control. 6-There was significant (p ≤ 0.05) decrease in sodium ions concentration in both UADX and UADX –DEX treated groups , however significant (p≤0.05) increased in potassium and calcium ions concentration were recorded during 15 days of treatment in UADX and UADX –Flud treated groups compared with control. 7- A significant (p ≤ 0.05) increase in serum total protein concentration and significant decrease in serum glucose concentration were recorded in UADX and UADX -Flud treated groups while a significant increased in glucose concentration was recorded in UADX-DEX treated groups during early periods of treatment compared with control. 8- A significant fluctuation in serum creatinine and urea concentrations were recorded in the UADX and UADX –Flud treated groups during early stage of treatment compared with control and UADX – DEX treated groups . 9- A significant (p ≤ 0.05) decrease in sperms characteristics were recorded in all treated groups compared with control. 10- A significant(p ≤ 0.05) increase in the relative weight of the thyroid and adrenal glands were recorded in UADX and UADX –Flud treated groups and a significant decrease in the relative weight of kidney and liver were observed in UADX and a significant decrease of testes relative weight was recorded in UADX and UADX –Flud compared with control. 11- Histopathological changes were found in left adrenal gland, thyroids gland, kidneys , liver and testes of UADX and UADX-Flud treated groups compared with control.

Second experiment This experiment was carried to elucidate the effects of long term treatment with Ketoconazol (KCZ) and Dexamethasone (DEX) for 60 days on the same parameters studied in the first experiment.

This experiment also include 24 local mal dogs ,they were divided into four equal groups 6 animals each as following : First group ( control) : The animals administration orally distal water for 60 days.

Second Group : The animals administration orally 10 mg / kg BW / day KCZ for 60 days.

Third group : The animal administrated orally 20 mg / kg BW / day KCZ for 60 days.

Fourth group : The animals administrated orally 0.5 mg / kg BW / day DEX for 60 days .

The result of this experiment revealed the following : 1- A significant (p ≤ 0.05) decrease in the body weight gains in all treated groups compared with control . 2- A significant (p ≤ 0.05) decrease in RBC count, Hb concentration and PCV value were found KCZ treated animals, while significant (p ≤ 0.05) increase in their value were recorded in DEX treated group at advanced periods of treatment . 3- A significant (p ≤ 0.05) decrease in the total WBC count, lymphocytes monocytes , and eosinophils percentages, on other hand significant increase in the neutrophils percentages were obsreved in all treated groups with advanced periods of treatment compared with control . 4-A significant(p ≤ 0.05) decrease in serum T3, T4, cortisol and testosterone hormones concentration were recorded in all treatments with advance periods of treatments, while significant (p ≤ 0.05) increase in serum ACTH concentration were recorded KCZ treated groups ,in contrast a significant(p ≤ 0.05) decrease in ACTH value was recorded in DEX treated group. 5- A significant (p ≤ 0.05) increase in the serum AST, and ALT enzymes concentration were recorded in all treated groups ,on other hand a significant (p ≤ 0.05) decrease in serum ALP enzyme concentration in KCZ treated groups , and a significant (p ≤ 0.05) increase in serum ALP value was recorded with advance periods of treatment in DEX treated animals. 6- A significant(p ≤ 0.05) decrease in serum total cholesterol concentration in KCZ treated groups and significant(p ≤ 0.05) increase in this value was recorded in DEX –treated group, whereas Significant (p ≤ 0.05) increase in serum triglyceride concentrations were recorded in all treatments with advance periods of treatments. 7- A significant (p ≤ 0.05) decrease in sodium ions concentration and a significant increase in potassium ions were recorded in KCZ treated groups while contrary results were recorded in DEX treated groups .Whereas a significant decrease in calcium concentration were recorded in all treated groups compared with control . 8- A significant (p ≤ 0.05) decrease in serum total protein concentration was recorded in all treated groups and a significantly decrease in serum glucose concentration was recorded in KCZ treated groups, while a significant increased value was recorded in the DEX treated groups with advanced periods of treatment . 9- A significant (p ≤ 0.05) increase in serum creatinine and urea concentrations were recorded KCZ treated groups, while significant(p≤0.05) decrease of these values were recorded in DEX treated group with advanced periods of treatment. 10- KCZ treated groups causes complete abolished of sperms characteristics, while Significant (p ≤ 0.05) decrease in sperms characteristics were recorded in DEX treated group . 11- A significant (p ≤ 0.05) increase of kidney, thyroid and adrenal gland relative weights.

Were recorded in 20 mg / kg Bw KCZ and 0.5 mg / kg BW DEX treated groups compared with control . 12- The treatment with KCZ and DEX causes atrophy and necrosis in ZG of adrenal gland, hyperplasia of thyroid glands ,degenerative changes in kidneys, periportal fibrosis and degenerative changes in the liver of KCZ and DEX treated groups .

Slight degenerative changes showed in the testes of animal treated with DEX and destructive changes found in the testes of animals treated with two doses of KCZ .
