الملخص الإنجليزي

-The present study has been designed in order to evaluate the Histological, Histochemical and Biochemical changes in the adrenal glands, kidneys and liver after long term treatment of dexamethasone in adult male rabbits, as a model for mammalian.

Thirty adult male rabbits were weighted and randomly divided into three groups: control and two treated groups A and B (10 rabbits in each group).

Control animals were orally administrated distilled water once daily.

Animals of group A and B were orally administrated with dexamethasone (0.2 and 1mg./ kg B.W.) once daily for 60.

All animals were weighted every 15 days up to end of the study.

As the final body weights of all animals were measured and blood samples obtained for biochemical evaluation.

Animals were sacrificed and samples taken from adrenal gland, kidney and liver.

Some of them use fresh to prepared the frozen sections and some other were obtained immediately for histopathological study.

The present study revealed the following results: A significant decrease in the body weight of groups A and B as compared with the control group (C).

A significant decrease in adrenal gland weight but a significant increase in kidney and liver weight in groups A and B as compared with the control group.

Biochemical study recorded a significant increase in serum cholesterol, T.G, HDL, LDL and VLDL concentration in Groups A and B compared with the control group.

A significant decrease in serum iron, Zinc, and Copper and serum calcium concentration of the treated Groups (A and B) as compared with control Group(C).

Histochemical examination of the treated Groups (A&B) show an increase in lipids droplets and increase adipocytes in the zones of cortex gloumerulosa & zona fasciculata.

In kidneys of the treated Group (B) sections showed increase accumulation of the lipids in the layers of convoluted tubules.

Liver Sections in the treated groups show an increase in the lipids in the hepatocytes and around the central.

Adrenal gland in the treated Group (B) shows increase in the fatty acid in the zona glomerulosa and zona fasciculata.

there is a slight increase in the Group (A) .

Kidney has been recorded a slightly increase in fatty acids level in the layers of the collecting tube in the sections of the treated Groups (A and B).

Liver Section of the treated groups (A&B) showed a mild increase in the fatty acids distribution in the hepatic cells near the central vein and bile ducts, as the fatty acids had a normal distribution in section of Control Group.

The Calcium salts in the adrenal gland are decreased in the Groups (A&B) sections in the cortex when compared with normal content of calcium salts as in the Control Group.

In kidney increase in calcium salts appearance between (A&B) treated groups compared with.

Liver histological examination showed a decrease the calcium salts in the lobules of liver in the section of treated Groups (A&B).

Copper salts in the Adrenal gland shows decrease in the appearance of copper in most part of adrenal gland cortex in the sections of Groups (A&B).

While Kidneys shows a low decrease in the copper in the glomeruli and collecting tubules in the Groups (A&B), while copper salts shows normal distribution in the Control group.

liver sections showed increase the copper salts in the hepatocytes of the treated groups (A&B).

Histopathological examination of the Adrenal gland cortex of the treated Groups (A & B) revealed the presence of Karyopyknosis, vacuolation the cytoplasm, congestions, edema, degeneration and necrosis in the in the cells of zona glomerulosa and zona fasciculata.

The sections of Kidneys of Group (A) showed present degeneration, necrosis and fibrosis in the epithelial lining of convoluted tubules, Section of Group (B) show Odem, fibrosis, bleeding and infiltration of inflammatory cells in the convoluted and collecting tubules, it is also found a shrinkage and the adhesion the Glomeruli with bowman's capsule.

The histological sections of Liver showed present of degeneration and necrosis of hepatocytes, appearance of inflammatory cells and congestion sinusoids and ballooning of cytoplasm found in the hepatocytes, concentric periductular fibrosis around bile ducts which leading eventually to destruction of affected bile ducts.

Many fields of liver histological sections revealed fatty changes.

It can be concluded that the daily oral administration of dexamethasone for 60 day leads to many disorders in lipids metabolism and distribution and causes a decrease in some trace elements in the body and had harmful effect on the tissues
