The present study was conducted at Veterinary Medicine College–University of Basra to investigate the effect of testosterone (Sustanon ® 250) abusing on some physiological, biochemical parameters, reproductive efficiency and histological changes in different organs.

The experiment was divided into three parts.

The first deals with physiological and biochemical parameters, whereas the second deals with the reproductive efficiency and the third deals with histological changes.

In the first part, 32 male and 32 female rats were used.

They were divided into four groups (each group consisted of 8 males and 8 females).

The first group served as control group in which the rats dosed by intramuscular injection with 10 μ l olive oil weekly for 12 weeks, the rest 3 treated groups (G1.

G2 and G3) in which the rats dosed by intramuscular injection with 50.

100 and 150 mg Sustanon ® 250 / kg B.

respectively weekly for 12 weeks , then all rats were terminated for the physiological and biochemical tests, the organs were removed for the histological part of the study .

The results revaled the following : The Sustanon ® 250 seems to reduce significantly the body weight gain in both sexes, the corrected weight mg / 100mg B.

of female rats were not affected as well as the liver corrected weight mg / 100mg B.

of male rats was significantly increased The Hb, PCV and RBC were significantly (p ≤ 0.05) increased in males and females by Sustanon ® 250 injection, but the WBC was not affected the reproductive hormones were significantly (p ≤ 0.05) altered in response to Sustanon ® 250 injection by elevation of testosterone in G1 and G2,whereas G3 was not affected.

The LH and FSH were significantly decreased (p ≤ 0.05) in all animals.

The Sustanon ® 250 through its effect on liver caused increasing both ALT and AST and decreasing the ALP and total serum protein in treated rats, whereas the muscle protein synthesis was significantly increased in treated male rats but was not affected in treated females.

The lipid profile was significantly affected by Sustanon ® 250 injection through elevation of TG and VLDL, but lowering the HDL, LDL and AI.

Sustanon ® 250 injection resulted in significantly elevated oxidative status of treated rats indicated by elevation of MDA level despite increased GSH concentration in blood , the uric acid level was significantly decreased(p ≤ 0.05) in all animals as a consequence of increased oxidative load.

The seminal analysis in response to Sustanon ® 250 injection revealed a significant reduction in the sperm concentration, viable sperm percentage and, morphologically normal sperm percentage and thereby, increased dead sperm percentage and, morphologically abnormal sperm percentage.

In the second part, 16 male and 32 female rats were used, from which 4 male and 8 female were dosed by intramuscular injection with 10 μ l olive oil weekly for 12 weeks, they are considered as control animals.

The rest rats were dosed by intramuscular injection with 150 mg Sustanon ® 250 / kg B.

Then they were divided into four groups : Group 1 consisted of 4 non treated males with 8 non treated females.

Group 2 consisted of 4 treated males with 8 treated females.

Group 3 consisted of 4 treated males with 8 non treated females.

Group 4 consisted of 4 non treated males with 8 treated females.

After they were allowed to mate for 12 weeks, the reproductive parameters were investigated which revealed the fertility percentage as 100 % in G1, whereas the female rats in G2, G3 and, G4 showed no pregnancy at all which indicated complete infertility.

The third part involved the investigation of histological changes in different organs (liver, kidney, heart, ovary, testes, prostate and, skeletal muscle) after 12 weeks of Sustanon ® 250 injection.
