Isolation and identification of pseudomonas aeruginosa and its ability in lectin production

This study was aimed to isolate lectin producing Pseudomonas aeruginosa from different clinical and environmental samples.

Ability of P.

aeruginosa isolates in lectin production and haemagglutination of RBCs for human blood groups (A, B, O) and sheep RBCs was studied.

Lectin produced by these isolates was partially purified by ammonium sulphate precipitation, dialysis, and gel filtration chromatography technique using Sepharose CL-6B.

First of all P.

aeruginosa was isolated from 34 of different serological (ear, blood, sputum, burns, cerebrospinal fluid and cystic fibrosis infections), and wastewater samples, from these samples, 18 bacterial isolate were obtained, 12 were identified as Pseudomonas spp.

according to their ability to grow on cetrimide agar medium, six out of them were identified as P.

aeruginosa according to physiological, morphological, and biochemical tests.

Results showed that all of these isolates has a high ability for lectin production, among them P.

aeruginosa S4 was selected for its high capacity in haemagglutination of different RBCs Results also showed that this isolate was a good producer for both PA-IL and PA-IIL lectin types.

After purification of PA-IL and PA-IIL lectins it was found that both of them was gave an inhibition of haemagglutination against different RBCs with specificity to galactose and mannose respectively.

The approximate molecular weight of the partially purified PA-IL and PA-IIL lectins were 57500 and 42700 Dalton respectively.
