Ciprofloxacin is a synthetic, broad spectrum antimicrobial agent, used for treatment of many microbial infections.

It is absorbed only from the region extending from the stomach to the jejunum (narrow absorption window), so most of the dose administered by conventional (immediate release) tablet is not absorbed.

For maximum absorption of this drug it was formulated in form of sustained release tablets that float over the gastric fluid for a long period of time. A formula of once daily 1gram ciprofloxacin given in United State patent (6162601) was selected to be a reference formula for this study.

It was claimed that the tablets of this formula are retained in the stomach for 5.5 hours and they release 35.49 % of their doses in the first hour and 98.72% after 6 hours (Talwaret al, 2001). The formula was formulated many times using different formulation methods to gain the stated results, but always the result of all formulations were fast complete disintegration of tablets and release high percentages of the active ingredient. It was found that the percentages of some excipients used in the formulation (viscolyzing agent, Poly Vinyl Pyrrolidone, and sodium bicarbonate) are insufficient to exert the needed floating behavior and release profile.

These percentages were modified to optimize the formula using full three level factorial design. The final result of this work was formulation of floating tablets, which stay floated for more than 6 hours and have a good release profile.
