Newcastle disease (ND) is a serious illness worldwide.

The disease was first isolated in the Sudan in the year 1964 (Karar and Mustafa, 1964). The disease caused very high economic losses in poultry industry.

The control of the disease was practiced by vaccination with komarov strain by intranasal route.

Although the vaccine was successfully controlled the disease but by the increase of poultry industry it was found difficult to use the intranasal route.

Nevertheless there are some reactions.

Being a mesogenic it was found that the vaccine induced some reactions shown by the appearance of some clinical ND signs.

Because of this problems trials for development of other vaccines originated from lentogenic strains started.

One of these methods is the administration of lentogenic vaccine strain through drinking water and spray. However, in our study a mesogenic vaccine strain was developed to substitute the milde lentogenic vaccine strain, and the way of demonstration of the mesogenic strain remain the same as the lentogenic vaccine.

The hot mesogenic strain was passaged many times in vero cell lines.

Then characterization of this vaccine strain was conducted to verify the chemical, Biological and culturing in cell and embrynated eggs and chicks.

So the vaccine was a result of plaquing in vero line designated as clone soba. Soba vaccine was used in drinking water, spray and nasal drops.

Chicks were vaccinated at age 7 days and three weeks.

The immune response was tested by (HI)_and ELISA.

After 2 weeks and one month post vaccination. So it was created new methods of vaccination with lentogenic strains via drinking water and spray. This study is to create a msesogenic vaccine strain from Komorov, by isolating strains by passage in vero cells.

After characterization to know chemicals, biological and culturing in cells and embrynated eggs and chicks.

So a vaccine was done from this clone. This vaccine was used in chicks of 7 day of age and three weeks and the immune antibodies were followed by using the three routes (Drinking water, spray and nasal drop).

The spray method was found to be the best.

At the end a new strain was found and compared with the available vaccines to immunity level and safety and simplicity.

The experiment was evaluated by HI and ELISA.

The cost was reduced according to this study.
