Taking into account the proteinic complexity of the Fasciola hepatica total antigens (FhTA), the exclusion-diffusion chromatography carried out on Sephadex G-100 gel, gave five antigenic fractions.
The SDS-PAGE electrophoresis, realized on the various antigenic components, made it possible to highlight several peptidic bands of varied molecular weights, ranging from 10 to 180 Kd.
The immunodiffusion test proved that the majority of FhTA fractions are endowed with antigenic activity, in the presence of rabbit antiserum.
The fraction (F.
1), separated starting from FhTA, is the only one to have developed a reaction of precipitation very positive with the Concanavaline A.
it is thus purified by ConA sepharose-4B affinity column.
The molecule obtained was then characterized by SDS-PAGE, and the electrophoretic profile revealed only one band of MW of approximately 40 Kd.
الملخص FRE
Compte tenu de la complexité protéique des antigènes totaux de Fasciola hepatica (FhTA), la chromatographie d’exclusion diffusion, réalisée sur gel sephadex G-100, a donné cinq fractions antigéniques.
L’électrophorèse SDS- PAGE, effectuée sur les différents constituants antigéniques, a permis de mettre en évidence plusieurs bandes peptidiques de poids moléculaires variés, compris entre 10 et 180 Kd.
Le test d’immunodiffusion prouve que la majorité des fractions de l’antigène FhTA sont douées d’activité antigénique, en présence de l’anti- sérum du lapin.
La fraction F.l, séparée à partir de l’homogénat total FhTA, est la seule fraction à avoir développée une réaction de précipitation très positive avec la Concanavaline A.
Elle est donc purifiée sur colonne d’affinité ConA- Sepharose 4B.
La molécule ainsi obtenue a été caractérisée par SDS- PAGE, et le profil électrophorétique a fait apparaître une seule bande de PM d’environ 40 Kd.
