Partial purification of prokallikrein from sheep pancreas

Sheep pancreatic prokaflikrein was partially purified by a sequential procedure of batch adsorption and anion exchange column chromatography, hydrophobic interaction chromatography and gel filtration chromatography.

The purification was monitored by measurement of the kininreleasing activity of the activated zymogen using kinin-radioimmunoassay.

The partially purified material (GF3) was isolated in a yield of 4.20/0 and purification fold of 41.8.

It showed 3 bands (on SDS-PAGE) that possibly represented isoenzymic forms of sheep pancreatic prokaflikrein with relative molecular weights corresponding to 45,000, 33.000 and 29,000.
