The present work focused on the biodegradation of polluted water with crude oil in the “South Baghdad Power Plant”.

Samples were collected from the heavy crude oil treatment unit.

Seventeen Gram negative and only one Gram positive bacteria were isolated.

Pseudomonas aeruginosa and Klebsiella pneumoniae were chosen to test their ability to biodegrade the heavy crude oil, results showed the ability of those isolates to biodegrade the heavy crude oil in solid medium with the addition of hydrocarbons.

These bacterial isolates which appeared good growth in the hydrocarbon media were identified using biochemical test and by VITEK 2 compact.

Results showed the efficiency of the two bacterial isolates to consume heavy crude oil, K.

pneumoniae recorded emulsion activity of 64.8 %, while it was 62.5 % for P.

aeruginosa.

Present results shows that the emulsions produced by bacterial isolates decrease the surface tension of the media to 44.50 and 43.30 Mn/m for P.

aeruginosa and K.

pneumoniae, respectively.

Results indicated that the optimum conditions for the growth of bacterial isolates and consuming hydrocarbons media were at optimum temperature 28 ºC, pH= 7 and incubation period of 28 days, were the percentage of hydrocarbons degradation were 78 % and 83 % for P.

aeruginosa and K.

pneumoniae, respectively.

The present results showed that a slight difference in the percentage of biodegradation heavy crude oil between concentrations of 1000 and 2000 mg/l for P.

aeruginosa which was 76.94 and 71.73 % respectively.

Summary II While K.

pneumoniae recorded lower percentage of consumption of 74.87 and 69.92 % respectively.

The efficiency of the isolated bacteria to degrade hydrocarbons was tested by Ultraviolet spectrophotometer and Infrared spectrophotometer and by following the changing in these spectrums since the beginning of the test till the end comparing with the control, results indicated that the two bacterial isolates have a good ability to degrade hydrocarbons.

the results of infrared spectrum showed a new absorption bands after 7 and 14 days of incubation in the wave length (2340-2362 cm -1), which refer to C-O group and accompanied by release of CO2 from the oxidation of hydrocarbons related to biodegradation process.

Also there is a changing in the absorption bands (1370-1460 cm-1) that correspond to C-H, C=C and aromatics C=O deformation due to formation of free fatty acids.

Moreover there is a significant decrease in the area of the peak at wave number (2850-2960 cm-1) that corresponding to aliphatic hydrocarbons stretching, with the appearance of a new absorption bands at wave number ( 3590-3726 cm-1) indication formation of hydroxyl group and hydrogen bonding.

The difference in absorption bands by its appearance and disappearance exhibited a good indication of the biodegradation of heavy crude oil
