Identification of Some bacterial pathogens associated with male infertility and detection of micro deletion in AZF genes on human Y chromosome using PCR technology

This study was aimed to investigate the relationship between the deletion of azoospermia factor (AZF) on Y chromosome and infected infertile male.

Three hundred infertile men were included in this study.

The men were divided in different categories according to the degree of infertility, 242 men (80.67%) with primary infertility and 58 men (19.33%) had secondary infertility with a history of azoospermia (35%), severoligozoospermia (9%), oligozoospermia (10%), asthenozoospermia (16%), oligoasthenozoospermia (30%), and 30 normal fertile men as a control group.

Anti-Chlamydia IgG, IgM and IgA antibodies were used to detect Chlamydia infection in semen.

It was done by using enzyme-linked immunosorbent assay (ELISA).

Nine cases (14.06%) of the primary infertile men had IgM, 44 (68.65%) had IgG, 7 (10.93%) had IgA, (IgM + IgA) were (0%), while 2(3.13%) had (IgG + IgA), and 2 (3.13%) had (IgG + IgM).

Chlamydia trachomatis was also detected serologically in secondary infertile men, it was found that 13 men (40.63%) had IgM, 14 (43.75%) had IgG, 2 (6.25%) had IgA, while (IgM + IgA) found in one case (3.12%), and no case with (IgG + IgA).

Two (6.25%) cases had (IgG + IgM).

Whereas 2 (6.66%) of control groups had IgM, 1 (3.34%) case had IgA.

In addition 27 (9%) of primary infertile men were infected with Neisseria gonorrhoeae , while 14 (4.6%) of secondary infertile men.

Escherichia coli isolated from one case (0.33%) with primary infertility, also one case (0.33%) with primary infertility had infection with Staphylococcus aureus and another case (0.33%) had the same infection showed positive result for this infection.

Treponema pallidum was detected in 3 (1%) primary infertile men, and in 2 (0.66%) men with secondary infertility.

Control group recoded (0%).

On the other hand, the primary and secondary infertility cases infected with Chlamydia trachomatis and Neisseria gonorrhoeae were genetically studied by DNA was extracted from peripheral blood, the STS (Sequence Tagged Site) primers tested on each subject were:SY86, SY84, SY81(AZFa); SY124, SY121 (AZFb);SY255, SY254, SY242, SY208 (AZFc); SY152, SY145, SY133 (AZFd); and Sy14 (SRY) as internal control.

It was found that 55(85.93%) of 64 (26.45) primary infertile men infected with Chlamydia trachomatis had Y-chromosome microdeletion (AZFa,b,c,d), and 24 (75%) of 32 (55.18%) with secondary infertility in comparison with control group that showed no microdeletions.

In addition men infected with Neisseria gonorrhoeae had Y-chromosome microdeletionin.

22 (81.48%) of 27 (11.16%) with primary infertility and 7(50%) of 14 (24.13%) with secondary infertility, in comparison with control group who showed no microdeletions.

Cases infected with Staphylococcus aureus, Escherichia coli and Treponema pallidum had no Y-chromosome microdeletion.
