Human Cytomegalovirus ( HCMV ) infection still represents the most common and potentially serious viral complication in pregnant women and Cytomegalovirus (CMV) is a major cause of abortion during pregnancy .

This study was amid to investigate the existence of HCMV in pregnant women, forty six blood specimens were collected from pregnant women (aged between 20 and 35 years old) which the specimens were suspected to be infected with Human Cytomegalovirus, were collected from Baghdad Teaching Hospital and Private Nursing Home Hospital During December 2011- to February 2012 .

Hence specimens were exanimated by using serological technique , Enzyme-linked Immunosorbent Assay (ELISA)( this method depends on the presence of antibodies against the virus in the patient’s serum.), this test rapid and useful for the primary diagnosis for the presence of HCMV in the specimens , According to the serological techniques, forty specimens were IgG and IgM positive and only six specimens were IgG positive and IgM negative.

II Molecular analysis was carried out for the detection of the presence of HCMV DNA in the specimens and that will confirm the results obtained from the serological test .

The results for the Real-Time PCR showed that ; 25 out of 46 (54.34%) patients were found to be HCMV positive while 21 out of 46 (45.65%) patients were found HCMV negative.

According to the results of this study, Real Time - PCR technique was used for the detection and quantification of Human Cytomegalovirus in serum specimens had high sensitivity ,effective and more specificity than serological methods ( ELISA).
