This study was aimed to characterization of genetic polymorphism between nine barley (Hordeum vulgare) varieties cultivated in Iraq using Random Amplified Polymorphic DNA (RAPD) markers.

To achieve this, the seeds were sown in a germination cabinet under controlled environment at 20°C and a 16/8 hrs.

light/darkness photoperiod.

Total genomic DNA of barley plant was extracted from two different sources, from 2-3 g of fresh leaves by using CTAB method, The average yields of DNA were in the range of 1090-8550 μg /g of leaf tissue with purity of 1.5-1.9, and also from dry seeds by using commercial kit, the DNA yield was 865-1035 μg/ mg of dry seed powder and a purity ranging between 1.4-1.6.

Molecular analysis was performed by using 18 random primers in (RAPD-PCR) technique for detection of genetic polymorphism between studied varieties.

These primers generated 177 amplified useful bands across nine varieties, 71 were monomorhpic bands 40% across all varieties and 106 were polymorphic bands 60% among the studied varieties.

polymorphic bands were observed ranging from 0-15 bands.

The primer OPD20 produced highest number of polymorphic bands 15 , while the primer OPA22 not produced any polymorphic bands.

the ratio of Polymorphism was 61.35% , primer OPE12 generated the highest percentage of polymorphism 100% compared with primer OPC12 generated lowest percentage of polymorphism 25% .

The size of the amplified fragments ranged from 4818 bp for primer OPH13 to 358 bp for primer OPC12.

It was also possible to find the DNA fingerprint of all studied barley varieties through the appearance of a number of bands that were II unique to each variety, such as IPA265 variety which distinguished by using the primers (GB7(1181)bp ), (S39(947)bp), and (A11(1958)bp).

IPA99 variety was differentiated by using the primers (S39(2334)bp) and (OPD2(1785)bp).

Tuwaitha variety was differentiated by using the primer (S39(1007)bp).

Hitra variety was distinguished by using the primers (S32(2318)bp), (OPG12(497)bp) and (OPC8(2140)bp).

Rayhan variety was differentiated by using the primers (S18(2435)bp)), (OPG12(2430)bp) and (OPC8(2200)bp).

Shuaa variety was differentiated by using the primer (OPE12(1814)bp).

Bawadi variety was distinguished by using the primers (GB7(1159)bp) and (OPF3(2377)bp).

Samir variety was differentiated by using the primers (S32(736)bp), (OPG12(2374)bp) and (OPD20(791), (709)and (448)bp).

AL_khair variety was differentiated by using the primers (S32(2242)bp), (S39(2440)bp), (A20(2401)bp), (OPD20(2445), (514) and (468)bp) and (OPC12(461) and (448)bp) respectively, that may be used in the future as cultivar specific marker.

Investigation of genetic distance using RAPD analysis showed that the lowest genetic distance was found between Samir and Bawadi varieties 0.13424 while the highest genetic distance was recorded between Samir and IPA265 varieties 0.4378.

Analysis of genetic relationship showed there were three major groups, the first included variety IPA265, the second included two subgroups, the first subgroup contained the variety IPA99 and the second subgroup contained the varieties Tuwaitha, Rayhan, Hitra and Shuaa.

The third included two subgroups, the first subgroup contained the varieties Bawadi and Samir, while the second subgroup contained the variety AL_khair.
