Chitosan as a natural biopolymer has been given focus in the current work as a tablet coating material for pharmaceutical applications.
For this purpose, three different molecular weights of chitosan were chosen; two were soluble in HCl (500 kDa, 143 kDa) and one was water soluble (13 kDa).
Experiments were carried out using three concentrations (1.5%, 1.0%, and 0.5% g/mL).
The effect of poly ethylene glycol (PEG) as a plasticizer was investigated using three different molecular weights (400, 4000, and 6000 Da) at concentrations of 10% and 20% w/w (of chitosan's weight).
Chitosan mixtures films were characterized by Moisture sorption study, Fourier Transform Infrared Spectroscopy (FTIR), XRay Powder diffraction (XRPD), Differential Scanning Calorimetry (DSC), Environmental Scanning Electron Microscopy (ESEM), viscosity and pH analysis for chitosan solutions mixtures, and finally tensile strength measurement.
The chitosan coating mixture " 1.5% w/v chitosan (500 kDa)/ PEG 400 (20% w/w) " was sprayed onto cimetidine tablets and dissolution of the tablets was carried out and compared with the uncoated tablets.
Results indicated that the two XV high molecular weights of chitosan were more hygroscopic than the lower molecular weight.
In addition, the PEG 400 rendered high hygroscopic character to all chitosans except for the 500 kDa, while PEG 4000 and 6000 had no effect on chitosan's hygroscopicity.
FTIR and DSC analysis indicated no interaction between the coating components and with the model drug.
XRPD indicated that all chitosan molecular weights (13 kDa, 143 kDa, and the 500 kDa) either without or with PEG 400 were showed as an amorphous state.
SEM indicated that the higher molecular of chitosan, the less porous the surface of chitosan films become.
Viscosity, pH analysis indicated that the impact of chitosan molecular weight on these values was significant with the highest molecular weight of chitosan (500 kDa) (p< 0.05).
From tensile strength analysis, it indicated that the impact of chitosan molecular weight on tensile force values was more obvious with the highest chitosan molecular weight.
Furthermore, PEG 400 showed better plasticizing efficiency than the higher M.wt PEGs (4000, 6000 Da).
Dissolution studies indicated no influence of the coating material on the dissolution profile of cimetidine, it was complied with USP pharmacopeia of cimetidine tablets
