Atherosclerosis is the principal cause of a constellation of cardiovascular diseases including coronary artery disease and abdominal aortic aneurysm.

Endothelial dysfunction, accumulation of cellular and extracellular substances in the arterial lumen and excessive vascular smooth muscle cell proliferation are the active players underlying the development of atherosclerotic inflammatory lesions.

All together, this events result in plaque formation, vascular remodeling and diminished oxygen supply.

Accordingly, considerable effort has been devoted to elucidate the mechanisms regulating VSM cell proliferation in this pathological condition.

In the present project, we test the involvement of Cytoglobin (Cygb); a novel globin type ubiquitously expressed in vertebrate tissues, in the regulation of excessive human aortic smooth muscle cell (HASMC) proliferation, one of the major steps characterizing the formation of atherosclerotic lesion within the aorta.

HASMCs were seeded at different cell densities and subjected to variable oxygen tensions and growth factors treatments.

Our results show an increase in Cygb expression at both mRNA and protein levels during hypoxia and this upregulation correlates with cell replication through increased rate of HASM cell proliferation, suggesting a possible role of Cygb in oxygen homeostasis by the regulation of cellular oxygen sensing and transport as a specific physiological adaptive mechanism to the lack of oxygenation in thickened aortic wall.
