In addition to p120 role in the stabilization of adherens junctions on the plasma membrane, p120 has been found in the cytoplasm where it can interact with Rho-GTPases, including RhoA, Cdc42, and Rac1.

In cadherin-deficient cells, all cellular p120 is stranded in the cytoplasm.

Abnormally high cytoplasmic levels of p120 associated with cadherin loss could directly dysregulate multiple events mediated by Rho-GTPases, such as cytoskeleton organization, motility, and gene transcription.

Previous studies suggested p120 is stable in the cytoplasm of E-cadherin deficient cells.

While investigating the effect of VE-cadherin down regulation on p120 level in Human Dermal Microvascular Endothelial Cells (HDMECs), we showed for the first time that p120 decreases in VE-cadherin deficient cells.

We also found that VE-cadherin loss relocalized p120, as p120 is no longer at cell borders.

This implies that VE-cadherin is necessary for the mobilization of p120 at cell borders and thereby maintaining the basal p120 level.

This led us to further study the mechanism by which VE-cadherin knockdown decreases p120 level.

We demonstrated that p120 decrease under VE-cadherin knockdown is not due to a transcriptional regulation.

Moreover, we reported that treatment of VE-cadherin deficient cells with ALLN or Lactacystin restored B-catenin but had no effect on p120 level.

This demonstrates that the proteosome, the calpain and cathepsins “L and B” are not involved in p120 degradation.

Further studies are required to determine the exact mechanism by which VE-cadherin knockdown decreases p120 cellular levels.
