Intimal hyperplasia is a major devastating pathological complication after angioplasty, bypass operations or stenting, and once begun, it is difficult to treat.

This neointimal formation induces a rapid smooth muscle cell (SMC) proliferation and migration that occurs as a result of the vascular smooth muscle (VSM) cells changing their phenotype from quiescent and differentiated to a synthetic phenotype.

Both cell proliferation and migration are modulated by changes in Ca2+ influx.

Part of the Ca2+-dependent actions are mediated through Ca2+/calmodulin-dependent serine/threonine kinases (CAMKs) including CAMKII.

VSM cells switch from a CAMKIIγ to a CAMKIIδ2 expression pattern along with their switch from the contractile to the synthetic phenotype which characterizes neointimal cells, suggesting that CAMKIIδ2 plays the role of intermediate between Ca2+ and downstream protein regulation responsible for the phenotypic switch in VSM cells.

CAMKIIδ2 physically interacts with Fyn, a member of the Src family of kinases (SFKs) which play a pivotal role in the regulation of many key cellular functions, including adhesion, migration, apoptosis, differentiation, cell cycle and organization of the cytoskeleton.

In this report we test the involvement of Fyn in VSM cell proliferation and migration which are hallmarks characterizing the switch from the quiescent to the synthetic phenotype proper to the neointimal VSM cells.

We found that Fyn plays different roles from those proper to the other SFKs; Fyn seems to be a positive regulator of cell adhesion and apoptosis and a negative regulator of cell migration.
