This laboratory study were performed to identify and characterize 2,4- DNT degrading bacterial strains isolated from historical explosive contamination soil in Maan quarries, southern Jordan.
The isolation and the identification by using morphological and biochemical characteristics obtain seven bacterial strains, all are monitored in BSM medium for ability to degrade 2,4-DNT as a sole carbon, nitrogen sources by using optical density (OD) for growth, 2,4-DNT disappearances and dropping of pH as indicator for nitrite releases in different concentrations, the results suggested that one strain of total seven bacterial have a high indicator for ability to degrade 2,4-DNT in 110μM concentration , this bacterial strain identification was Pseudomonas oryzihabitans.
Results from the biodegradation experiments by Pseudomonas oryzihabitans indicate that the biodegradation rates of 2,4-DNT with this strain were highly dependent on environmental conditions such as pH and temperature, with optimal conditions obtained at 37 °C and pH ≈7.5.
Supplemental nitrogen and carbon sources in BSM medium are also tested, and the results showed that the maximum degradation rate obtained when BSM medium contain 0.2% (w/v) yeast extract or 0.2% (w/v) glucose respectively, all carbon and nitrogen sources are used except L-proline increase the degradation percentage after ~40 hours of incubation.
However, the BSM with 0.2% glucose or 0.2% succinate as carbon sources have been the highest initial degradation rate respectively, and also the BSM with 0.2% L-serine or 0.2% casein as nitrogen source have been the highest initial degradation rate respectively.
