The use of Doppler broadening technique (DBT) for studying the changes in the microstructure of different materials, subjected to different effects, is currently a very active field in Physics, Chemistry, and Materials Science.

However, there are no attempts to employ this technique for biological systems.

Therefore, in this work, the use of DBT applied in biology has been suggested in order to provide a new non-destructive method to monitor the microstructural changes in biological tissues due to many effects.

Doppler-broadening spectrometer has been established using a high pure germanium (HpGe) detector of 2.4 keV resolution at 1.33 MeV of 60Co.

Factors affecting the performance of the present spectrometer are experimentally investigated.

The RESOLUTION program is employed in order to correct the lineshape spectra from various contributions, mainly, the background and Compton continuum contributions.

Then the correction spectra were analyzed to central (S) and wing (W) shape parameters.

The measurements of S and W parameters are carried out on different animal tissues under various conditions, such as, change of fixation solutions, their concentrations and aging time, change of temperature, and these parameters are also studied with tumour tissues.

The results of fixation experiment allude to that the S parameter is very sensitive to changes in tissues due to their fixation in formalin at concentration (3.5%, 5%, 7.5%, 10%), alcohol (ethanol, methanol)at concentration (50%, 100%), and normal saline.

The results demonstrate the potential usefulness of the Doppler broadening method for finding the optimum type and time required for fixation tissues.

The R parameter, which is the indication for o-Ps formation probability, and S parameter have been investigated in the tissues exposed to different temperatures.

The values of S and R parameters are varied with respect to temperatures which are associated with the phase transition of water inside the cells.

Lineshape parameters (S and W) in normal and abnormal tissues (tumours) in mice’s mammary adenocarcinoma have also been determined as a function of tumour growth.

It is found that S parameter is considerably changed with the distribution of voids which are affected as the tumour is developed.

The results suggest that DBT can successfully be employed to distinguish between normal and abnormal tissues.
