Back ground Greater attention has been paid to the traditional medical practices in general and herbal medicine in particular even in developed countries.

This study aimed to screen some of the traditionally used medicinal plants for their potential contraction and relaxation effect on small intestine in rabbits.

Methods Five plants claimed in folk medicine to have a contraction or relaxation effect on small intestine.

These are: Allium porrum, Nerium oleander, Cassia senna, Ammi visnaga, and Foeniculum vulgare.

They were investigated for their contraction and relaxation effect on jejunum of rabbit by using organ bath in vitro.

In order to establish the dose-response curve for the plant extracts, six male rabbits were used for each experiment.

Further pharmacological studies were performed for elucidation of the effect of most potent plant which caused a contraction or a relaxation effect on the intestinal contraction.

Pharmacological studies for the best contraction plant were made to compare between intestinal contraction after addition of the plant extract alone as agonist and intestinal contraction after addition of both antagonist drugs and the plant extract.

While pharmacological study for the relaxation plant was compared between intestinal Abstract contraction after the addition of an agonist drug alone and that after addition of both relaxation plant as antagonist followed by the addition of agonist drugs.

Results Allium porrum, Nerium oleander, Cassia senna, showed a statistically significant contraction effect except Nerium oleander which caused a non significant effect.

Ammi visnaga, Foeniculum vulgare showed a statistically significant relaxation effect in a dose dependent manner.

The most potent plant extract that caused a contraction was Allium porrum, while the most potent one that caused a relaxation was Ammi visnaga.

Conclusion Increased of intestinal contraction by Allium porrum, Nerium oleander, and Cassia senna and reduced one by Ammi visnaga, and Foeniculum vulgare were found in the present study.

Atropine, adrenaline, and chlorpheniramine reduced the effect of Allium porrum on intestinal contraction, and produce shift to the right of the dose response curve.

Neostigmine and pilocarpine effects were inhibited by administration of Ammi visnaga.

Neostigmine and pilocarpine caused a shift to the right of the dose response curve.

This effect of neostigmine is more potent than the pilocarpine and this may be due to tissue selectivity of neostigmine on the muscarnic receptors of the jejunum.
