Leishmania parasite is the causative agent of different clinical diseases.

Fetal calf serum (FCS) is the most expensive and main part ingredient of Leishmania cultures.

In this study the efficacies of different concentrations (10%, 15%, 20%, 25% and 30%) of the filtered healthy and diabetic human urine were evaluated as a growth stimulator of Leishmania in culture media as alternative to the FCS.

The results showed that culture media enriched with the filtered healthy human urine in concentration 25% supported the parasite growth significantly (p<0.05) of the growth rate of parasites compare with all studied concentrations.

While according to gender, healthy males urine at 30% gave significantly (p<0.05) the highest growth rate of parasites, whereas healthy females urine at 25% gave the significant (p<0.05) highest growth rate of parasites.

On the other hand, the results confirmed that culture media without FCS enriched with filtered healthy human urine in concentration of 30% gave a significant (p<0.05) highest growth rate of parasites.

While according to gender, 25% of healthy males urine gave a significant (p<0.05) highest growth rate of parasites, whereas healthy females urine at 30% concentration gave a significant (p<0.05) highest growth rate of parasites in M199 media without FCS.

So healthy females urine at 30% can be use as alternative to the FCS in M199 media.

The results of diabetic human urine, showed that the higher growth rate of parasites was scored significantly (p<0.05) in concentration of 25%.

While according to gender, diabetic males urine at 25% gave a significant (p<0.05) highest growth rate of parasites, whereas in diabetic females urine at 30% gave a significant (p<0.05) highest growth rate of parasites.

In addition, the studied of culture media without FCS enriched with diabetic human urine, the results showed a significant (p<0.05) higher growth rate of parasites in concentration of 25%.

While according to gender, diabetic males urine at 25% gave significantly (p<0.05) highest growth rate of parasites, whereas in diabetic females urine at 30% gave a significant (p<0.05) highest growth rate of parasites.

Finally, female healthy urine scored significantly more growth rate of parasites than the male and total urine (healthy, diabetic and control) and may use as alternative instead of FCS.

The disease outcome in mice injected with the best concentration of urine(healthy and diabetic) that obtained best parasite growth as well as parasites without urine, was not influenced by the type of media used in this study, all animals infected with parasites maintained in media with or without urine whether it contains FCS or not showed lesions that developed greatly and hastily, and there were all died with an obvious eroded noses after two weeks of infection.
